**Purpose of Use:** This kit is designed for the quantitative determination of anti-tuberculosis antibodies in human serum, plasma, and other related biological samples. It provides a reliable method for detecting immune responses against tuberculosis, supporting early diagnosis and monitoring of treatment efficacy.
**Experimental Principle:** The assay is based on the double-antigen sandwich immunoassay technique. A solid-phase antigen is pre-coated onto the microwells of a microplate. After incubation with the sample, anti-tuberculosis antibodies present in the specimen bind to the immobilized antigens. Subsequently, HRP-labeled antigens are added, forming a complex of antigen-antibody-enzyme-labeled antigen. Following a thorough wash step, TMB substrate is introduced, which changes color under the catalytic action of HRP. The reaction is stopped by an acidic solution, resulting in a yellow color. The intensity of the color is directly proportional to the concentration of anti-tuberculosis antibodies in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the antibody concentration is calculated from a standard curve.
