Leucorrhea sample pretreatment

1. Collect samples such as cervical swabs, and rinse them with 2–3 ml of normal saline. After allowing the sample to rest for 30 minutes, carefully remove the supernatant, leaving a thin layer of sediment at the bottom. Then add an equal volume of normal saline, mix well, and either prepare a smear or centrifuge the sample. Ensure all steps are performed clearly and precisely. 2. Before centrifugation, make sure to remove any centrifuge tube sleeves from the centrifuge. Balance the rotor by ensuring the pointer is centered. Select centrifuge tubes of similar weight and place the sample tubes in the rotor. Balance the tubes using physiological saline if needed. Position the balanced tubes symmetrically—typically placing them in the inner ring first (6 holes in the inner ring, 6 in the outer ring). Centrifuge at 500 rpm for 3–5 minutes. If a complete pellet is required, increase the speed to 3000–3500 rpm and centrifuge for 5 minutes. In this experiment, however, 500 rpm for 3 minutes was sufficient. 3. After centrifugation, carefully remove the supernatant. A small amount may be left behind if necessary. Mix the remaining suspension thoroughly, then proceed to prepare a smear directly from the sample or aspirate it. If all the supernatant is removed, add an equal volume of normal saline, mix again, and proceed with the smear. 4. Apply 3–5 µl of the sample onto a slide using a pipette tip, spread it into a circular area of approximately 1 cm². Label the slide clearly with the sample number and relevant information. Make sure to mark both sides so that the label does not get smudged during handling. 5. Once the smear is mostly dry, examine it under a microscope to check for cleanliness and uniformity. If there is still excess mucus, repeat the washing and centrifugation process to obtain a clearer sample. 6. Fix the smear with 95% ethanol for 2–3 minutes, then wash it with distilled water. This step helps remove excess ethanol and rehydrates the sample, making it easier for staining agents to penetrate. 7.
