Human diseases are often caused by disordered regulation of gene expression. MicroRNA (miRNA) is an important gene expression regulator that has been widely concerned in recent years, and plays an important role in the occurrence and development of human diseases. The research team led by Professor Shen Nan integrates the clinical advantages of the Department of Rheumatology of Renji Hospital Affiliated to Shanghai Jiaotong University and the basic research strength of the Institute of Health Sciences, Shanghai Academy of Life Sciences, Chinese Academy of Sciences. Recently, Arthritis Rheum., The most influential journal in the field of rheumatology, published the latest research results on miRNAs involved in the expression and regulation of lupus inflammatory factors: miR-125a can directly negatively regulate the transcription factor klf13 of T cells to promote RANTES Expression, thereby reducing the level of inflammatory chemokine RANTES produced by T cells. The level of miR-125a in peripheral blood T cells of lupus patients was significantly lower than that of normal people. After the activation of T cells in patients, the expression levels of KLF13 and RANTES were also significantly higher than those in the normal control group. When overexpression of miR-125a in T cells of patients, the expression of KLF13 and RANTES can be reduced during this process.
These studies suggest that the defect of miR-125a expression in lupus patients may be one of the reasons for the abnormally elevated RANTES levels in vivo. MiR-125a may serve as a new drug intervention target, and targeted intervention of miR-125a expression level may develop into new lupus. Treatment.
