Breakthrough in research and development of hand-foot-mouth disease genetic engineering vaccine of Chinese Academy of Sciences

Under the guidance of Researcher Huang Zhong, Liu Qingwei, a PhD student of the Vaccine and Antiviral Strategy Research Group of the Shanghai Pasteur Institute of the Chinese Academy of Sciences, developed a genetically engineered vaccine for hand-foot-mouth disease of Coxsackie virus A16 (CA16). This is the first international attempt to develop a CA16 genetically engineered vaccine. The results show that the VLP (virus-like particle) -based CA16 vaccine strategy is feasible. It is a bivalent hand and mouth for the further development of anti-EV71 (Enterovirus 71) and CA16. The disease vaccine laid the foundation. Researchers used the insect baculovirus expression system to co-express the P1 and 3CD proteins of CA16 to obtain virus-like particles of CA16. The particles are similar in structure to the virus, contain no viral nucleic acid, and are not infectious. After immunizing mice with VLP, CA16-specific serum IgG antibodies were produced. The in vitro neutralization test showed that VLP immune serum can effectively inhibit the infection of homologous and heterologous CA16 viruses. Further animal challenge experiments have shown that anti-VLP serum has a good protective effect against lethal doses of homologous or heterologous CA16 virus challenge.
